Adenovirus (ADV) is a non-enveloped linear double-stranded DNA virus with a diameter of approximately 90-100 nm and has the ability to infect a wide range of cells and tissues. ADV is mainly used as vectors to deliver related proteins or antigens, such as for the development and commercial production of COVID-19 vector vaccines. In addition, they are also used as vaccines against a variety of tumor-related antigens. ADV is generally cultured in HEK293 cells and released from the cells after lysis. After the cells are opened, a large amount of DNA will be released into the supernatant. Generally, nucleases are usually added to degrade DNA and other invalid components. The virus harvests liquid after lytic enzymatic digestion needs to be clarified and filtered to enter the downstream purification process steps. This report mainly introduces the successful cases of Cobetter Depth Filtration products for clarification and filtration of Adv26 harvest liquid.

Experimental Conditions

For the culture medium with an initial cell density of 1×106 cells/ml and 6.67x106 cells/ml, the materials are lysed under the same conditions, and the depth filtration product code 0140PC (23cm2, 0.04-9um) is used for clarification and filtration. Control the inlet flow rate to 100-150LMH for P-max filtration. The final operating pressure is controlled at 1.0 bar, and the mixed turbidity of the filtrate is controlled within 100NTU.
